Durante il loro lavoro, i ricercatori hanno sintetizzato una copia del genoma di Mycoplasma mycoides, che \u00e8 stata poi impiantata in una cellula ospite, diventando completamente vitale e mantenendo la capacit\u00e0 di auto-replicarsi.<\/p>\n<p>La nuova ricerca prosegue il lavoro di Craig. Se in precedenza gli scienziati avevano creato un modello digitale del DNA di un organismo reale e sintetizzato una molecola sulla base di esso, il nuovo progetto va oltre, utilizzando il codice originale del DNA. Gli scienziati lo hanno notevolmente rielaborato prima di sintetizzarlo e testarne la funzionalit\u00e0.<\/p>\n<p>I ricercatori hanno iniziato con il genoma originale di C. crescentus, che contiene 4000 geni. Come in tutti gli organismi viventi, la maggior parte di questi geni non porta alcuna informazione ed \u00e8 considerata &laquo;DNA spazzatura&raquo;. Dopo un'analisi, i ricercatori hanno concluso che solo circa 680 di essi erano necessari per mantenere in vita il batterio in laboratorio.<\/p>\n<p>Dopo aver rimosso il &laquo;DNA spazzatura&raquo; e ottenuto il genoma minimo di C. crescentus, il team ha continuato il lavoro. Il DNA degli organismi viventi \u00e8 caratterizzato dalla presenza di ridondanza incorporata, in cui la sintesi della stessa proteina \u00e8 codificata da diversi geni in pi\u00f9 punti della catena. I ricercatori hanno sostituito oltre 1\/6 delle 800.000 lettere di DNA durante l'ottimizzazione per rimuovere il codice duplicato.<\/p>\n<p>&laquo;Grazie al nostro algoritmo abbiamo completamente riscritto il genoma in una nuova sequenza di lettere di DNA, che non assomiglia pi\u00f9 all'originale&raquo;, afferma Bit Christen, co-autore dello studio. &laquo;In questo modo, la funzione biologica a livello di sintesi proteica \u00e8 rimasta invariata&raquo;.<\/p>\n<p>Per verificare se la catena ottenuta avrebbe funzionato correttamente in una cellula vivente, i ricercatori hanno coltivato un ceppo di batteri che conteneva nel proprio DNA sia il genoma naturale di Caulobacter che segmenti del genoma artificiale. Gli scienziati hanno disattivato singoli geni naturali e testato la capacit\u00e0 dei loro analoghi artificiali di svolgere lo stesso ruolo biologico. Il risultato \u00e8 stato piuttosto impressionante: circa 580 dei 680 geni artificiali si sono rivelati funzionanti.<\/p>\n<p>&laquo;Grazie alle conoscenze acquisite, saremo in grado di migliorare il nostro algoritmo e sviluppare una nuova versione del genoma 3.0&raquo;, afferma Christen. &laquo;Riteniamo che nel prossimo futuro saremo in grado di creare cellule batteriche viventi con un genoma completamente sintetico&raquo;.<\/p>\n<p>Nella fase iniziale, tali ricerche aiuteranno i geneticisti a verificare la correttezza delle loro conoscenze sulla comprensione del DNA e sul ruolo dei singoli geni, poich\u00e9 qualsiasi errore nella sintesi della catena porter\u00e0 alla morte o all'incompletezza dell'organismo con il nuovo genoma. In futuro, per\u00f2, porteranno alla creazione di micro-organismi sintetici, progettati per compiti specifici.
